In some cases, another plugin could be interfering with your site functionality, including uploads. Is there a fix for that one? Re-activate NextGEN Gallery only and try uploading again. After downloading it just unzip it and then upload it in the "wp-content/uploads" and then in the Slideshow option give the path of it as "wp-content/uploads/imagerotator.swf".
